The four teams comprise Jahna Boshoff, Marion le Roux and Bridgette Johnson; Sibongile Mpakanyiswa, Monica Villalobos and Tanya Weyers; Julie Farrington, Alicia Proud and Monique Hansen; and Petra Schuller, Ingrid Bohme and Tanya Tiefenthaler.
Enya Fehler and Andrea Doefert will drive in a fifth vehicle as representatives of Africa Adventure to provide logistical support and carry out administrative duties.
The teams were selected while 18 women from all over the world attended a workshop in Bitou which culminated in the participants choosing those who would take on the challenge.
Each person had to write down who they would like in their team and also propose the members of the other three teams. The final 12 were chosen from these lists.
Event organiser Rachel MacRae said: "It has been fantastic to see the tourism sector pulling together to make this expedition happen. This is a non-profit initiative, so sponsorship is essential to us.
"Although it is the women who are making the journey, we feel the whole industry is a part of it."
